LAESI-MSI


LAESI-MSI services from Protea provide a superior sample introduction method that significantly reduces your sample preparation and offers you in situ mass spectrometric analysis: a more exact profile of where and how your molecules of interest reside in your sample. LAESI-MSI services are available for a wide variety of sample types and investigation modalities.

2D LAESI-MSI
Tissue samples are mounted on standard glass slides and analyzed using the LAESI DP-1000 System connected to either a Thermo Scientific Q Exactive™ Hybrid Quadrupole-Orbitrap mass spectrometer, a high resolution Waters Synapt® G2 HDMS mass spectrometer, or a Thermo Scientific LTQ Velos™ linear ion trap mass spectrometer. Analysis is typically performed on samples with the target area of up to 10mm x 10mm at the standard 200μm spatial resolution (or a different target area and spatial resolution within a specified range to be discussed with customer). Bacterial colonies are analyzed directly on standard growth medium plates, and cell cultures are analyzed directly after pelleting. Standard data reports contain up to five contour plots of desired m/z of interest, as well as well as representative MS fingerprints from the entire sample and/or select regions. Further contour plots and MS/MS confirmation can also be performed at an additional cost.

3D LAESI-MSI
Plant tissue samples (of up to 1mm thick) are mounted on standard glass slides and analyzed using a LAESI DP-1000 System connected to either a Thermo Scientific Q Exactive™ Hybrid Quadrupole-Orbitrap, a Thermo Scientific LTQ Velos™, or a high resolution Waters Synapt® G2 HDMS mass spectrometer. Analysis is typically performed on samples with the target area of up to 10mm x 10mm at 200µm spatial resolution (or larger if analyzing large samples, as discussed with customer). Depth profiling is performed by targeting the laser at the same location with multiple laser pulses. Each laser pulse provides one layer of information from the sample. Pulse depth varies between sample types with averages between 20 and 40µm per pulse. Standard data reports contain up to five contour plots of desired m/z of interest, as well as representative MS fingerprints from the entire sample and/or select regions. Sample size, number of layers, and number of masses to plot define the price. Further contour plots and MS/MS confirmation can also be performed at an additional cost. For mammalian tissue analysis, serial tissue sections are analyzed independently and then reconstructed into 3D molecular images.
