Choosing PAGE Gel Concentration for Resolving a Protein of Interest

Polyacrylamide gels can be formed with varying pore sizes for a wide range of protein separation. The size of the pores is dependent on the amount of total acrylamide per unit volume and the percentage of cross-linkers available. The smaller pore size (higher percent acrylamide gel, e.g. 15%) is beneficial for separation of low molecular weight proteins and the larger pore size (lower percent acrylamide gel, e.g. 5%) is beneficial for separation of high molecular weight proteins.
